Melasma is a common chronic dermatosis, which predominantly affects females. Its characterized by patches and spots usually in areas exposed to UV light. Melasma can occur in different profiles of skin, from light brown to darker skin tones and it negatively impacts the quality of life. The formation of melasma depends on melanogenesis, which is a physiological process of production of melanin. The modulation of melanogenis is a key procedure used in cosmetic and pharmaceutical products to control skin pigmentation disorders. For a centuries, the control of skin hyperpigmentation disorders have therapeutically been a challenge for cosmeceutical and pharmaceutical industries, however, in the last decades, studies have significantly increased on tyrosinase function, melanogenesis and melasma production and melanin biosynthesis processes, therefore, new strategies for the treatment of skin hyperpigmentation disorders have been designed based on tyrosinase inhibition as the key enzyme in melanin biosynthesis and melasma production. The use of topical therapies, combination therapies, chemical peels, microdermabrasion, microneedling, lasers, oral agents, botanical agents and intravenous agents are some of the common approaches for the treatment of melasma. The growing demand for such products, is projected to drive the market growth for the next decades and is expected to be worth for $23.8 billion by 2027. Kojic acid has shown to be effective in the tyrosinase inhibition, the enzyme behind melanin biosynthesis, however, it is unstable in a long range of conditions such as light and pH. Thus, to overcome this fact, the acrylic acid and KA derivatives raised as a potent and promising strategy. Acrylic acid and KA derivatives were successfully synthesized and their affinity values for the tyrosinase was calculated, as compounds DFA and DAK showed high affinity for the tyrosinase, suggesting to be effective inhibitors. However, inhibitory assay and ADMET studies for the compounds is necessary.
